1. Lack of Process Monitoring
Common dyes like bromophenol blue in traditional buffers do not offer clear visibility of the electrophoresis front, especially in thick gels or fast electrophoresis runs, making it difficult to monitor the progress accurately.
2. Inability to Assess Transfer Efficiency
Standard dyes cannot be transferred onto membranes, leaving the transfer process unmonitored until antibody incubation, preventing early-stage quality control.
3. Limited Loading Flexibility
Traditional 2X buffers limit the volume of sample that can be loaded into each well, often requiring concentration or reducing sample volume for low-concentration samples, thus hindering detection of low-abundance proteins.
4. Dye Compatibility Issues
