Profiling ADP binding proteins in complex samples is difficult, and the main challenges include:
1. State Specific Detection Is Hard
Traditional antibody based or biochemical assays rarely measure ADP bound state directly, making it difficult to distinguish ADP engaged proteins from total protein abundance.
2. No Proteome Wide Co-Capture
In complex proteomes it is difficult to unbiasedly capture multiple ADP binding proteins and their binding competent states at the same time, limiting network level comparisons across conditions.
3. Inhibitor and Ligand Profiling Is Low Throughput

Desthiobiotin-ADP is an ADP analog that carries a desthiobiotin handle through a reactive linkage. After endogenous nucleotides are reduced, it selectively forms a covalent adduct at conserved active site lysine residues in the nucleotide binding pocket, directly tagging functional ADP binding sites.
